Maternal continuous oral oxycodone self-administration alters pup affective/social communication but not spatial learning or sensory-motor function

The broad use/misuse of prescription opioids during pregnancy has resulted in a surge infants with Neonatal Opioid Withdrawal Syndrome (NOWS). Short-term irritability and neurological complications are its hallmarks, but the long-term consequences unknown. A newly-developed preclinical model oxycodone self-administration enables adult female rats to drink (?10/mg/kg/day) before pregnancy, after delivery, maintain normal liquid intake, titrate dosing, avoid withdrawal. Oxycodone was detected serum mothers pups. Growth parameters dams pups litter mass size were similar controls. There no differences paw retraction latency thermal stimulus between Control at postnatal (PN) 2 or PN14. had motor coordination, cliff avoidance, righting time, pivoting, olfactory spatial learning from PN3 through PN13. Separation-induced ultrasonic vocalizations PN8 revealed higher call frequency relative (p<0.031; Cohen’s d=1.026). Finally, displayed withdrawal behaviors (p’s<0.029; d’s>0.806), males only vocalized more than first minute testing (p’s<0.050; d’s>.866). Significant effects corroborated by estimation plots. Our rat oral shows exacerbated affect/social communication sex-dependent manner spared cognition sensory-motor behaviors. This reproduces selective aspects human opioid use enabling longitudinal analysis how maternal changes emotional behavior offspring.
